| 保存條件 | Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C. |

background: ERAS belongs to the small GTPase superfamily. Ras family. Ras proteins bind GDP/GTP and possess intrinsic GTPase activity. The ERas gene is expressed in embryonic stem (ES) cells and promotes their in vitro proliferation and tumorigenicity. ERas-null ES cells maintain pluripotency but show significantly reduced growth and tumorigenicity, which are rescued by expression of ERas complementary DNA or by activated phosphatidylinositol-3-OH kinase. Function: Ras proteins bind GDP/GTP and possess intrinsic GTPase activity. Plays an important role in the tumor-like growth properties of embryonic stem cells (By similarity). Subunit: Interacts with PIK3CD (By similarity). Subcellular Location: Cell membrane; Lipid-anchor; Cytoplasmic side. Similarity: Belongs to the small GTPase superfamily. Ras family. Paraformaldehyde-fixed, paraffin embedded (Mouse embryo); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Incubation: Anti-ERAS Antibody, conjugated (bs-4950R-FITC) 1:400, 1.5 hours at 37°C; DAPI (5ug/ml, blue, C-0033) was used to stain the cell nuclei.
Blank control (Black line): U937(Black).
Primary Antibody (green line): Rabbit Anti-ERAS/FITC antibody (bs-4950R-FITC)
Dilution: 1μg /10^6 cells;
Isotype Control Antibody (orange line): Rabbit IgG-FITC .
Protocol
The cells were fixed with 4% PFA (10min at room temperature)and then permeabilized with 90% ice-cold methanol for 20 min at room temperature. The cells were then incubated in 5%BSA to block non-specific protein-protein interactions for 30 min at room temperature .Cells stained with Primary Antibody for 30 min at room temperature. Acquisition of 20,000 events was performed.
