保存條件 Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.
產(chǎn)品介紹 background:
Amylin is commonly found in pancreatic islets of patients suffering diabetes mellitus type II, or harboring an insulinoma. While the assosciation of amylin with the development of type II diabetes has been known for some time, a direct causative role for amylin has been harder to establish. Recent results suggest that amylin, like the related beta amyloid (Abeta) assosciated with Alzheimer's disease, can induce apoptotic cell death in particular cultured cells, an effect that may be relevant to the development of type II diabetes.

Function:
Selectively inhibits insulin-stimulated glucose utilization and glycogen deposition in muscle, while not affecting adipocyte glucose metabolism.

Subunit:
Interacts with IDE and INS. Can form homodimers. Interaction with INS inhibits homodimerization and fibril formation.

Subcellular Location:
Secreted.

Post-translational modifications:
Amyloid fibrils are degraded by IDE.

Similarity:
Belongs to the calcitonin family.

胰島淀粉樣肽(islet amyloid polypeptide,IAPP)又稱amylin 或糖尿病相關(guān)肽是由胰腺β細(xì)胞產(chǎn)生的含37個氨基酸的一種短肽。IAPP降低胰島素的敏感性,抑制肌肉組織中葡萄糖的攝取和糖原的合成,抑制病人攝取食物。
據(jù)報(bào)道胰腺癌病人血漿IAPP濃度顯著升高,而在胃癌、結(jié)腸癌、肝癌、肺癌中正常,在糖尿病人中正常或稍低。由于血漿IAPP濃度上升是胰腺癌的一個早期特征,因而檢測IAPP或IAPP釋放因子對于胰腺癌的早期診斷是有價(jià)值的。但目前還不清楚IAPP釋放因子的生化特性。
胰島淀粉樣多肽又稱胰淀素 IAPP ,胰島淀粉樣多肽在胰島β細(xì)胞中合成。并在β細(xì)胞中分泌,在各種疾病因素的誘導(dǎo)和激發(fā)作用下,IAPP生成異常增加而代謝降低,從而使其在胰島中聚集沉淀。
過量沉積的IAPP使胰島β細(xì)胞膜和胰島β細(xì)胞的超微結(jié)構(gòu)破壞,從而抑制了胰島素的分泌和對糖代謝平衡的協(xié)調(diào)作用,同時(shí)也抑制了葡萄糖對胰島素分泌的刺激作用,從而導(dǎo)致了胰島素分泌的不足,糖代謝平衡失調(diào),引發(fā)了糖尿病,胰島淀粉多肽是引發(fā)糖尿病的根本原因。由此可見如果能消除過量的IAPP,胰島β細(xì)胞的功能就可以恢復(fù)正常,糖尿病就有可能治愈。
