PHLDA1 is involved in regulation of apoptosis induced by signaling of T cell receptor in murine lympocytes. As any potent regulator with pleiotropic effects related to its overexpression, PHLDA attracts attention in both cell biology research and human tissue pathology. Function: Seems to be involved in regulation of apoptosis. May be involved in detachment-mediated programmed cell death. May mediate apoptosis during neuronal development. May be involved in regulation of anti-apoptotic effects of IGF1. May be involved in translational regulation. Subunit: Interacts with RPL14, EIF3S7 and PABPC4. Subcellular Location: Cytoplasm. Cytoplasmic vesicle membrane. Nucleus, nucleolus. Note=Colocalizes with intracellular vesicles. Tissue Specificity: Widely expressed with highest levels in pancreas. Strongly expressed by benign melanocytic nevi, and progressively reduced expressed in primary and metastatic melanomas (at protein level). Similarity: Contains 1 PH domain. Secondary: IRDye800CW Goat Anti-Rabbit IgG at 1/20000 dilution

Paraformaldehyde-fixed, paraffin embedded (rat brain tissue); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (PHLDA1) Polyclonal Antibody, Unconjugated (bs-1364R) at 1:400 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0023) instructionsand DAB staining.
Tissue/cell: mouse pancreas tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
Antigen retrieval: citrate buffer ( 0.01M, pH 6.0 ), Boiling bathing for 15min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30min; Blocking buffer (normal goat serum,C-0005) at 37℃ for 20 min;
Incubation: Anti-PHLDA1/TDAG51 Polyclonal Antibody, Unconjugated(bs-1364R) 1:200, overnight at 4°C, followed by conjugation to the secondary antibody(SP-0023) and DAB(C-0010) staining
